Can't find direct answer using search so....
To add functionality to a website homepage, I need to change the file name from index.htm to default.asp
Considering it is a reasonably well Google-ranked site, will this change be to its detriment? I have done a link: check which shows the same amount of inbound links to www. domain .com as it does to www. domain .com/index.htm, so does it follow that changing to www. domain .com/default.asp will be okay?

You cannot change your real homepage. Your home page is www.yourdomain.com and NOT www.yourdomain.com/whatever.html
All your links should be going to www.yourdomain.com and you should never link internally to www.yourdomain.com/index.html or anything else.
So, your home page filename really should not matter whatsoever. If there are links to a filename then get rid of them immediately.

To future-proof a site, always omit the index file filename itself from any links. Let the server find and display the index page. The user does not need to know its real name.
